CJC-1295 is a synthetic peptide modeled on growth hormone-releasing hormone, the hypothalamic signal that prompts the pituitary to release growth hormone. Its sequence corresponds to the first twenty-nine residues of human GHRH, with four substitutions that slow enzymatic breakdown. Early descriptions placed the compound in research on growth hormone deficiency and related conditions, and later literature groups it with the long-acting GHRH analogs. The name appears in both laboratory and popular fitness writing, where it sometimes labels chemically different peptides.
Two related peptides circulate under the CJC-1295 label, and they differ mainly in how long they persist in circulation. The version carrying a drug affinity complex includes a maleimidopropionic acid linker that forms a covalent bond with serum albumin. The other version, usually written as modified GRF(1-29) or tetrasubstituted GRF(1-29), lacks that linker and is cleared quickly. Mixing the two produces inconsistent readings of published half-life values, because the linker rather than the receptor-facing sequence drives most of the difference.
The core sequence keeps the receptor-binding region of GHRH while replacing four positions that are vulnerable to dipeptidyl peptidase-4 and other proteases. Substitutions at positions 2, 8, 15, and 27 raise metabolic stability relative to the natural hormone. The N-terminal residues remain essential for activity, so changes there generally lower potency. Molecular weight sits near 3368 daltons for the tetrasubstituted analog without the linker, while the albumin-binding form is heavier because of the added maleimide group.
Clearance profiles diverge sharply between the two versions. The albumin-binding molecule stays in plasma for several days, whereas the unmodified analog is largely gone within about half an hour in reported work. Cleavage by dipeptidyl peptidase IV is a major contributor to the short life of the unmodified sequence. These gaps mean the two versions cannot be substituted for each other in study design or in reading results side by side.
Reports on this compound commonly follow serum growth hormone and insulin-like growth factor 1 across defined time windows. Protocols differ in sampling frequency, assay platform, and participant characteristics, which makes direct comparison between publications difficult. Some work focuses on pulsatile release patterns instead of average concentrations. Whether repeated exposure alters endogenous hormone rhythms over long periods remains an open question, and the formal literature is thinner than the volume of informal commentary implies.

Identity and purity are established with reversed phase high performance liquid chromatography coupled to mass spectrometry. The chromatographic step separates the target peptide from truncated sequences and deletion products, while the mass measurement confirms the expected molecular weight to within a fraction of a dalton. Because the two common variants differ by the presence of the linker, mass alone can distinguish them in the unconjugated state. Amino acid analysis and peptide mapping are used when sequence level confirmation is required.
Lyophilized material is generally stable for extended periods when held at minus twenty degrees Celsius or below and protected from moisture and light. In solution the peptide is more labile; bond hydrolysis, aggregation and oxidation of susceptible residues all proceed faster at ambient temperature. Repeated freeze and thaw cycles should be avoided because they promote clumping and loss of soluble material. The conjugated variant adds a further consideration, since the maleimide group can hydrolyze in aqueous buffer and lose its ability to react with albumin.
Laboratory handling centers on minimizing exposure to water, heat and oxygen before use. Working solutions are typically prepared in sterile water or a mild buffer, and any residual particulate matter is removed by filtration. When the powder dissolves slowly, a small proportion of acetonitrile or dilute acetic acid is sometimes added as a co-solvent. Containers are kept sealed and desiccated between uses. Records of lot number, reconstitution date and storage conditions support later comparison of results across experiments.
The drug affinity complex is a maleimidopropionic acid group attached to a lysine side chain. It reacts with the free thiol of cysteine-34 on circulating albumin, forming a covalent bond. This conjugation keeps the peptide in the bloodstream and shields it from rapid renal filtration and proteolysis. Reported circulation half-lives for the albumin-bound form fall in the range of roughly six to nine days in early human studies.
Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
CJC-1295 is a synthetic analogue of growth hormone-releasing hormone, built on the 29-amino-acid GHRH(1-29) fragment. Four substitutions distinguish it from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes reduce enzymatic cleavage and extend the peptide's persistence in circulation. The compound is discussed in two forms, one carrying a drug affinity complex and one without it.
=== Human Therapeutics === In recent years, gene and cell therapy therapeutic options have become increasingly present in the clinic. For some of these therapeutics, the administration of the drug directly into the central nervous system is optimal for the treatment of neurological disorders, while avoiding a severe immune response. Additionally, most of the dose is introduced directly into the target area with ICV injection. In addition to these therapies, ICV injection has been used for the delivery of chemotherapies, treatment of carcinomatous meningitis, and other neurological disorders. In the design of gene therapies, the proper adeno-associated virus (AAV) serotype must be selected. AAV is effective at transporting genetic material in vivo, and there are more than 100 serotypes for AAV that have been identified. Each serotype has a different binding capacity to cell surface receptors. Three serotypes have been identified for their promising specificity to the central nervous system. In a 2017 study, AAV2/1, AAVDJ8, and AAV9 were administered to neonatal mice via ICV injection. The brains of these mice were analyzed for GFP expression following this procedure. The results of this showed that AAV2/1 had higher expression in the cortical layers while penetrating less to the midbrain compared to the AAVDJ8 and AAV9 serotypes. The results indicate that ICV injection of AAV vectors is successful for having a lasting expression of the transgene.
=== Degradation of DNA during bisulfite treatment === A major challenge in bisulfite sequencing is the degradation of DNA that takes place concurrently with the conversion. The conditions necessary for complete conversion, such as long incubation times, elevated temperature, and high bisulfite concentration, can lead to the degradation of about 90% of the incubated DNA. Given that the starting amount of DNA is often limited, such extensive degradation can be problematic. The degradation occurs as depurinations resulting in random strand breaks. Therefore, the longer the desired PCR amplicon, the more limited the number of intact template molecules will likely be. This could lead to the failure of the PCR amplification, or the loss of quantitatively accurate information on methylation levels resulting from the limited sampling of template molecules. Thus, it is important to assess the amount of DNA degradation resulting from the reaction conditions employed, and consider how this will affect the desired amplicon. Techniques can also be used to minimize DNA degradation, such as cycling the incubation temperature. In 2020, New England Biolabs developed NEBNext Enzymatic Methyl-seq, an alternative enzymatic approach to minimize DNA damage. Instead of bisulfite, APOBEC is used to convert C into U. Distinction between C, 5mC, and 5hmC is granted by the further modifications that "protect" the modified bases from APOBEC.
The chemical energy stored in ATP (the bond of its third phosphate group to the rest of the molecule can be broken, allowing more stable products to form, thereby releasing energy for use by the cell) can then be used to drive processes requiring energy, including biosynthesis, locomotion, or transportation of molecules across cell membranes.
=== Police withdrawal === Swelled by thousands of new recruits and an increasingly sophisticated arsenal of heavy weapons, PLAN undertook more direct confrontations with the security forces in 1973. Insurgent activity took the form of ambushes and selective target attacks, particularly in the Caprivi near the Zambian border. On the evening of 26 January 1973 a heavily armed group of about 50 PLAN insurgents attacked a police base at Singalamwe, Caprivi with mortars, machine guns, and a single tube, man portable rocket launcher. The police were ill-equipped to repel the attack and the base soon caught fire due to the initial rocket bombardment, which incapacitated both the senior officer and his second in command. This marked the beginning of a new phase of the South African Border War in which the scope and intensity of PLAN raids were greatly increased. By the end of 1973, PLAN's insurgency had engulfed six regions: Caprivi, Ovamboland, Kaokoland, and Kavangoland. It also had successfully recruited another 2,400 Ovambo and 600 Lozi guerrillas. PLAN reports from late 1973 indicate that the militants planned to open up two new fronts in central South West Africa and carry out acts of urban insurrection in Windhoek, Walvis Bay, and other major urban centres. Until 1973, the South African Border War was perceived as a matter of law enforcement rather than a military conflict, reflecting a trend among Anglophone Commonwealth states to regard police as the principal force in the suppression of insurgencies.

== Inactivation == The study and manipulation of viruses, regardless of application, often requires a preceding inactivation step. The goal is to render the virus safe and non-infectious while preserving sufficient structural integrity to allow meaningful analysis. For serological studies, the preservation of surface epitopes is critical, as these protruding viral structures mediate antibody binding. Antigenicity is particularly important in vaccine development, where efficacy depends on eliciting a significant host immune response—especially the production of neutralizing antibodies. In contrast, for molecular studies the preservation of viral genetic material is prioritized, such as RNA or DNA, rather than surface structures. This requirement is central to many microbiological diagnostic methods, including PCR, other amplification techniques, and sequencing. Virus inactivation can be achieved through physical or chemical methods, and often a combination of approaches is employed. This is especially necessary when working with highly pathogenic viruses, where complete sterility is essential to eliminate the risk of exposure. Physical inactivation methods include heat (pasteurization), ultraviolet (UV) light, gamma radiation and pH-alteration inactivation. Chemical approaches encompass guanidinium-based chaotropic salts, detergents (e.g., sodium dodecyl sulfate [SDS], methanol, Tween compounds, Triton X-100), β-propiolactone, hydrogen peroxide, and aldehydes (e.g., formaldehyde, paraformaldehyde, glutaraldehyde), as well as aromatic disulfides.
Initially, Valve used a hierarchical structure typical of other development firms, driven by the nature of physical game releases through publishers that required tasks to be completed by deadlines. However, as Valve became its own publisher via Steam, it found the hierarchical structure was hindering progress. After completing Half-Life 2, Valve transitioned to a flat organization. Outside of executive management, Valve does not have bosses, and uses an open allocation system. Valve's marketing manager, Doug Lombardi, said: "Nobody writes a design doc and hands it to somebody and says, 'you go build this'. It's the teams that are coming up with the ideas and pushing in the directions that they want to take the product." This approach allows employees to work on whatever interests them, but requires them to take ownership of their product and mistakes they may make, according to Newell. Newell recognized that this structure works well for some but that "there are plenty of great developers for whom this is a terrible place to work". Following the difficult development of Half-Life 2, Newell said he became "obsessed" with improving Valve's work-life balance. Although Valve has no bosses, some employees hold more influence due to seniority or relationships. De facto project leads became "centralized conduits" for organization and sharing information, and decisions are made collectively. Valve uses a process named Overwatch to gather feedback from senior members, which teams may use or ignore. The success of Steam means that Valve is not dependent on the success of its games.

Natural GHRH is degraded quickly by dipeptidyl peptidase-4 and related enzymes, giving it a half-life measured in minutes. CJC-1295 carries substitutions that slow that breakdown, so it stays intact longer. Both act at the same pituitary receptor and produce the same class of signal.
The label was first attached to the albumin-binding form during early development work. A shorter analog without the linker later became known by the same name in informal use. Published papers normally state which version was studied, so the methods section resolves the ambiguity.
It is a synthetic peptide rather than a steroid, and it does not occur naturally in the body. Its sequence is derived from a hypothalamic hormone called growth hormone-releasing hormone. It works as a receptor agonist rather than as a replacement for that hormone.
It acts on the growth hormone-releasing hormone receptor found on pituitary somatotroph cells. Activation of that receptor triggers growth hormone release through a cyclic AMP dependent pathway.
