A practical reference on Albumin conjugation: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.

Both forms act at the pituitary receptor for growth hormone-releasing hormone and increase growth hormone output, which in turn raises insulin-like growth factor 1. A long-acting analog produces sustained rather than pulsatile stimulation, and the physiological consequences of that pattern are not fully settled. Published human data on the extended form remain limited, and much of what circulates in discussion traces to early company reports rather than independent replication. How sustained exposure affects normal feedback remains an open question.
The distinguishing feature of the DAC form is a maleimide-containing group that reacts with the free thiol of cysteine-34 on human serum albumin. This reaction forms a covalent bond without enzymatic assistance, and it takes place after the peptide enters the bloodstream. Because albumin is abundant and long-lived, the attached peptide is carried through circulation far longer than an unmodified fragment would survive. The chemistry is a deliberate pharmacokinetic strategy rather than a change to receptor activity.
Enzymatic protection is a separate mechanism from plasma protein binding. The four substitutions in the backbone reduce recognition by dipeptidyl peptidase IV, which normally cleaves the natural hormone within minutes. Without the reactive group, this resistance still yields only a short window of activity, generally reported in the range of tens of minutes. With it, reported half-lives in early human work extended to several days. The size of that gap is the main practical distinction between the two materials.
The drug affinity complex is a maleimidopropionic acid group attached to a lysine side chain. It reacts with the free thiol of cysteine-34 on circulating albumin, forming a covalent bond. This conjugation keeps the peptide in the bloodstream and shields it from rapid renal filtration and proteolysis. Reported circulation half-lives for the albumin-bound form fall in the range of roughly six to nine days in early human studies.
Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
| Property | Value | Notes |
|---|---|---|
| Duration with linker | Several days | Reported in early human work |
| Duration without linker | Tens of minutes | Short plasma residence |
| Albumin attachment site | Cysteine-34 | Covalent maleimide reaction |
| Primary receptor | Pituitary GHRH receptor | Stimulates growth hormone release |
| Downstream marker | Insulin-like growth factor 1 | Indirect measure of activity |
Two forms circulate under the CJC-1295 name, and they differ by a single appended group. The version without a drug affinity complex carries four substitutions along the peptide chain, including a D-alanine near the amino terminus and replacements at three other positions. These changes block the enzyme dipeptidyl peptidase IV and remove a methionine residue that is prone to oxidation. The modified fragment is frequently labeled MOD GRF 1-29. Naming conventions are inconsistent across informal sources, which is a common source of confusion.
The second form adds a maleimide-bearing linker to the lysine at the carboxyl end. This group reacts with cysteine-34 on circulating serum albumin, forming a covalent bond that keeps the peptide in the bloodstream for far longer. ConjuChem developed the molecule as a way to extend the action of a peptide without frequent administration. The albumin attachment is the defining structural feature of the drug affinity complex version. Whether continuous exposure produces effects distinct from shorter pulses remains an unresolved research question.
Receptor level activity follows the canonical GHRH pathway. The peptide binds the GHRH receptor, a class B G protein coupled receptor on somatotroph cells of the anterior pituitary. Binding raises intracellular cyclic AMP, which promotes calcium entry and the release of growth hormone into the bloodstream. Because the molecule acts at the same receptor as the endogenous hormone, its effect is superimposed on the natural pulsatile rhythm rather than replacing it. Whether sustained receptor occupation leads to desensitization is not fully settled.
CJC-1295 belongs to a class of synthetic peptides modeled on growth hormone releasing hormone, a forty-four amino acid signal produced by the hypothalamus. The compound is built from the first twenty-nine residues of the natural sequence, with four substitutions introduced at positions 2, 8, 15 and 27. These changes were designed to slow enzymatic breakdown while preserving receptor activation. The result is a peptide that is shorter than native GHRH and considerably more resistant to ordinary clearance pathways in circulation.
==== Tissue regeneration ==== To repair and regenerate damaged tissue, nanogels have been explored to not only encapsulate drugs and growth factors for local administration, but also to serve as porous scaffolds at a tissue implantation site. Boron-containing temperature-responsive nanogels formed a solid scaffold upon injection into a critical bone defect and continued to induce the production of new osteoblast cells. To treat the effects of myocardial infarction, one in vivo study loaded temperature-responsive nanogels with cardiac stem cells and observed improved cardiac function through an increase in left ventricular ejection. Blood vessels have been successfully regenerated in an in vivo model of ischemia using nanogels to encapsulate vascular endothelial growth factors. Heparin-based nanogels loaded with growth factors have also been tested in the regeneration of the urethral muscle that causes urinary incontinence.
== Video games == Games created during this time period often used a motif of nuclear war, as was the threat at that time. Some of the games listed have been made after the conclusion of the Cold War, but feature a central plot point around the Cold War.

Similarly, the Food Packet, Survival, Abandon Ship (FPSAS) and Food Packet, Survival, Aircraft, Life Raft (FPSALR) are fitted into the storage areas on lifeboats. The "Jimmy Dean", a pre-packaged shelf-stable ration containing, among other items, a pre-made Jimmy Dean brand deli-style sandwich, is often issued in the field to U.S. servicemen as an alternative to MREs.
=== Basal rate determination === Basal insulin requirements will vary between individuals and periods of the day. The basal rate for a particular time period is determined by fasting while periodically evaluating the blood sugar level. Neither food nor bolus insulin must be taken for 4 hours before or during the evaluation period. If the blood sugar level changes dramatically during evaluation, then the basal rate can be adjusted to increase or decrease insulin delivery to keep the blood sugar level approximately steady. For instance, to determine an individual's morning basal requirement, they must skip breakfast. On waking, they would test their blood glucose level periodically until lunch. Changes in blood glucose level are compensated with adjustments in the morning basal rate. The process is repeated over several days, varying the fasting period, until a 24-hour basal profile has been built up which keeps fasting blood sugar levels relatively steady. Once the basal rate is matched to the fasting basal insulin need, the pump user will then gain the flexibility to skip or postpone meals such as sleeping late on the weekends or working overtime on a weekday. Many factors can change insulin requirements and require an adjustment to the basal rate:

=== Breast cancer === Flutamide was studied for the treatment of advanced breast cancer in two phase II clinical trials but was found to be ineffective. Out of a total of 47 patients, only three short-term responses occurred. However, the patients in the studies were selected irrespective of AR, ERTooltip estrogen receptor, PRTooltip progesterone receptor, or HER2 status, which were all unknown.
The fur is soft, with dense underfur and long, coarse, thick guard hairs. The fur is not as shaggy or coarse as that of brown bears. American black bear skins can be distinguished from those of Asian black bears by the lack of a white blaze on the chest and hairier footpads. Despite their name, black bears show a great deal of color variation. Individual coat colors can range from white, blonde, cinnamon, light brown or dark chocolate brown to jet black, with many intermediate variations existing. Silvery-gray American black bears with a blue luster (found mostly on the flanks) occur along a portion of coastal Alaska and British Columbia. White to cream-colored American black bears occur in the coastal islands and the adjacent mainland of southwestern British Columbia. Albino individuals have also been recorded. Black coats tend to predominate in humid areas, such as Maine, New England, New York, Tennessee, Michigan and western Washington. Approximately 70% of all American black bears are black, though only 50% in the Rocky Mountains are black. Many in northwestern North America are cinnamon, blonde or light brown in color and thus may sometimes be mistaken for grizzly bears. Grizzly (and other types of brown) bears can be distinguished by their shoulder hump, larger size and broader, more concave skull. In his book The Great Bear Almanac, Gary Brown summarized the predominance of black or brown/blonde specimens by location:
In compression fractures of the vertebrae, paresthesia is a complication due to the inflammation of the tissue surrounding nerves. Organs commonly affected by AS, other than the axial spine and other joints, are the heart, lungs, eyes, colon, and kidneys. Other complications are aortic regurgitation, Achilles tendinitis, AV node block, and amyloidosis. Owing to lung fibrosis, chest X-rays may show apical fibrosis, while pulmonary function testing may reveal a restrictive lung defect. Very rare complications involve neurologic conditions such as the cauda equina syndrome.
In these places, the bricks would become a blank canvas for artists, intellectuals and young people to tackle the "Wall" phenomenon. To commemorate the 20th anniversary of the fall of the Berlin Wall, the 3D online virtual world Twinity reconstructed a true-to-scale section of the Wall in virtual Berlin. The MTV Europe Music Awards, on 5 November, had U2 and Tokio Hotel perform songs dedicated to and about the Berlin Wall. U2 performed at the Brandenburg Gate, and Tokio Hotel performed "World Behind My Wall". Palestinians in the town of Kalandia, West Bank, pulled down parts of the Israeli West Bank barrier, in a demonstration marking the 20th anniversary of the fall of the Berlin Wall. The International Spy Museum in Washington D.C., hosted a Trabant car rally where 20 Trabants gathered in recognition of the 20th anniversary of the fall of the Berlin Wall. Rides were raffled every half-hour and a Trabant crashed through a Berlin Wall mock up. The Trabant was the East German people's car that many used to leave DDR after the collapse. The Allied Museum in the Dahlem district of Berlin hosted a number of events to mark the 20th anniversary of the fall of the Berlin Wall. The museum held a Special Exhibition entitled "Wall Patrol – The Western Powers and the Berlin Wall 1961–1990" which focused on the daily patrols deployed by the Western powers to observe the situation along the Berlin Wall and the fortifications on the GDR border. === Environmental === Buildup of cadmium levels in the water, air, and soil has been occurring particularly in industrial areas. Environmental exposure to cadmium has been particularly problematic in Japan where many people have consumed rice that was grown in cadmium-contaminated irrigation water. This phenomenon is known as itai-itai disease. People who live near hazardous waste sites or factories that release cadmium into the air have the potential for exposure to cadmium in air. However, numerous state and federal regulations in the United States control the amount of cadmium that can be released to the air from waste sites and incinerators so that properly regulated sites are not hazardous. The general population and people living near hazardous waste sites may be exposed to cadmium in contaminated food, dust, or water from unregulated or accidental releases. Numerous regulations and use of pollution controls are enforced to prevent such releases. Some sources of phosphate in fertilizers contain cadmium in amounts of up to 100 mg/kg, which can lead to an increase in the concentration of cadmium in soil (for example in New Zealand).

Plutonium recovered from LWR spent fuel, while not weapons grade, can be used to produce nuclear weapons at all levels of sophistication, though in simple designs it may produce only a fizzle yield. Weapons made with reactor-grade plutonium would require special cooling to keep them in storage and ready for use. A 1962 test at the U.S. Nevada National Security Site (then known as the Nevada Proving Grounds) used non-weapons-grade plutonium produced in a Magnox reactor in the United Kingdom. The plutonium used was provided to the United States under the 1958 US–UK Mutual Defence Agreement. Its isotopic composition has not been disclosed, other than the description reactor grade, and it has not been disclosed which definition was used in describing the material this way. The plutonium was apparently sourced from the Magnox reactors at Calder Hall or Chapelcross. The content of Pu-239 in material used for the 1962 test was not disclosed, but has been inferred to have been at least 85%, much higher than typical spent fuel from currently operating reactors. Occasionally, low-burnup spent fuel has been produced by a commercial LWR when an incident such as a fuel cladding failure has required early refuelling. If the period of irradiation has been sufficiently short, this spent fuel could be reprocessed to produce weapons grade plutonium.
Following Sigmund Freud's pioneering work, ideas stemming from psychoanalytic theory also began to take root in psychiatry. The psychoanalytic theory became popular among psychiatrists because it allowed the patients to be treated in private practices instead of warehoused in asylums.

== Pathophysiology == Episcleritis is caused by inflammation due to the activation of immune cells, including lymphocytes and macrophages. In most cases, the cause of episcleritis is never determined (idiopathic). An identifiable cause is discovered in about one third of cases. Several diseases are associated with episcleritis, including systemic vasculitis (polyarteritis nodosa, granulomatosis with polyangiitis, Behçet's disease), connective tissue diseases (rheumatoid arthritis, relapsing polychondritis, systemic lupus erythematosus), psoriatic arthritis, ankylosing spondylitis, Cogan syndrome, rosacea, gout, atopy, Crohn's disease, and ulcerative colitis. 59 percent of patients with relapsing polychondritis have either episcleritis or scleritis. Rarely, episcleritis may be caused by scleritis. Very rarely, episcleritis is associated with infections, including Lyme disease, tuberculosis, syphilis, and herpes zoster. The redness in the eye associated with episcleritis is due to engorgement of the large episcleral blood vessels, which run in a radial direction from the limbus. Typically, there is no uveitis, or thickening of the sclera.
==== Design principles ==== Practical applications of MOFs for hydrogen storage are met with several challenges. For hydrogen adsorption near room temperature, the hydrogen binding energy would need to be increased considerably. Several classes of MOFs have been explored, including carboxylate-based MOFs, heterocyclic azolate-based MOFs, metal-cyanide MOFs, and covalent organic frameworks. Carboxylate-based MOFs have by far received the most attention because

A maleimide moiety reacts with the thiol of cysteine-34, forming a covalent bond. The reaction occurs in circulation without enzymatic catalysis.
The group is intended to alter distribution and persistence, not receptor engagement. The peptide portion still binds the pituitary receptor, so the primary difference is duration rather than potency.
Not firmly. Early reports describe several days for the extended form, but independent confirmations are sparse, and values vary with assay method and study design.
The two share the same 29-amino-acid backbone. CJC-1295 carries an additional albumin-binding moiety that markedly extends its residence time in circulation, while the form without that moiety clears faster. Literature sometimes applies the name loosely to either variant.
